Question

How can the effect of pH on the rate at which amylase digests starch be investigated?

Answer · five matched stages
  1. 1

Begin by placing one drop of iodine solution into each well of a clean spotting tile, ready for repeated tests.

  2. 2

Measure fixed volumes of amylase solution, starch solution and a buffer at the chosen pH. Keep their concentrations unchanged, bring them to a constant temperature, mix them and start the timer immediately.

  3. 3

At 30-second intervals, remove a fresh sample of the reaction mixture and add it to the next iodine drop.

  4. 4

Continue until a sample leaves the iodine orange-brown, showing that no starch remains. Record the time, then repeat with buffers at other pH values while keeping temperature, volumes and concentrations constant.

  5. 5

Compare the recorded times, or calculate the reaction rate as one divided by the time taken.

Question

How can the effect of pH on the rate at which amylase digests starch be investigated?

Answer · five matched stages
  1. 1

Place iodine drops in a spotting tile.

  2. 2

Mix fixed volumes and concentrations of amylase, starch and a chosen pH buffer; keep the mixture at constant temperature and start timing.

  3. 3

Every 30 seconds, add a sample to fresh iodine.

  4. 4

Record when the iodine stays orange-brown, then repeat at other pH values while keeping other variables constant.

  5. 5

Compare the times, or calculate rate as 1/t.

Simple ideas should not be harder to remember than necessary.

Aymro grew from using revision resources that often turned one clear idea into a long card or mixed in content from the wrong course.

Built from real revision experience, it keeps each card focused and each revision set relevant.
